人血清对生殖支原体LAMPs诱导THP-1细胞表达TNF-α的影响 被引量:1

摘要 目的探讨人血清对生殖支原体(Mg)脂质相关膜蛋白(LAMPs)诱导TNF-α表达的影响。方法提取对数期Mg的LAMPs,分别加入脂蛋白脂肪酶,蛋白酶K及人血清预处理后,刺激THP-1细胞,ELISA法测定其TNF-α水平。结果 Mg LAMPs为一混合蛋白,其活性成分为脂质成分,能激活THP-1细胞表达大量TNF-α,且与LAMPs浓度呈明显的剂量依赖性。人血清能上调LAMPs诱导的TNF-α水平,且与LAMPs浓度呈明显的剂量依赖性。结论人血清上调Mg LAMPs诱导THP-1细胞表达TNF-α。 Objective To investigate the effects of human serum on TNF-α expression activated by lipid-associated membrane proteins ( LAMPs) of Mycoplasma genitalium( Mg) . Methods LAMPs were extracted and pretreated with lipo-protein lipase or protease K or serum human. THP-1 cells were stimulated and the expression of TNF-α was detected by ELISA. Results LAMPs are mixed proteins and the active ingredients are lipid moiety. The secretion of TNF-αwas media-ted in LAMPs induced THP-1 cells and was significantly increased by LAMPs in a dose-dependent manner. TNF-αlevels were significantly up-regulated in LAMPs activated THP-1 cells by Human serum and was significantly increased by LAMPs in a dose-dependent manner. Conclusions The human serum raised TNF-αlevels by Mg LAMPs induced THP-1 cells.
