卡替诺尔和氟西汀对映体的高效毛细管电泳分离 被引量:7

摘要 考察了以羧甲基_β_环糊精 (CM_β_CD)、β_环糊精 (β_CD)、羟丙基_β_环糊精 (HP_β_CD)、二甲基_β_环糊精 (DM_β_CD)为手性选择剂 ,在50mmol/L醋酸三乙胺缓冲溶液中分离卡替诺尔和氟西汀对映体。该文还通过考察手性选择剂的浓度、背景电解质的酸度、背景电解质的类型等因素对对映体手性分离的影响 ,对分离条件进行了优化 ,初步探讨了手性识别机理。实验结果表明 :用约4mmol/L的CM_β_CD分离氟西汀和卡替诺尔对映体 ,能使对映体达到良好分离,不仅节约了分析成本 ,也简化了分析过程。 A method for separating enantiomers of fluoxetine and carteolol is described by HPCE using β _ cyclodextrin(β _ CD), carboxylmethyl _ β _ cyclodextrin(CM _ β _ CD), hydroxypropyl _ β _ cyclodextrin(HP_ β _ CD) and dimethyl _ β _ cyclodextrin(DM _ β _ CD) as a chiral selector. The separations were performed on an uncoated capillary of 50 cm×75 μm. The effects of types and concentrations of CDs in buffer on chiral resolution were investigated. The effects of pH and types of the background electrolytes on chiral resolution were also examined and separation conditions were optimized. It was observed that a good separation of carteolol or fluoxetine can be achieved by using approx 4 mmol/L of CM _ β _ CD, which not only saves analysis cost, but also simplifies the procedure. The mechanism of the chiral resolution was discussed briefly.
