三七总皂苷诱导U937细胞凋亡及对Caspase-3表达的影响 被引量:1

摘要 目的观察三七总皂苷(PNS)对人急性单核细胞白血病细胞系U937细胞的增殖、凋亡及Caspase-3表达的影响。方法用不同浓度PNS作用于U937细胞,采用MTT法检测细胞增殖活性,用流式细胞术检测细胞周期和凋亡,Western-blot法检测Caspase-3蛋白表达。结果 PNS抑制U943细胞生长并呈时间和剂量依赖性;分别予100、200、400、800mg/L处理U937细胞48h,细胞凋亡率分别为(5.93±0.15)%、(7.43±0.70)%、(12.3±0.38)%和(20.4±0.91)%,对照组细胞凋亡率为(1.73±1.50)%(P<0.01),并诱导细胞G1期阻滞;Western-blot结果显示,PNS使Caspase-3蛋白表达上调。结论 PNS能抑制U937细胞增殖,并可能通过上调Caspase-3蛋白表达诱导细胞凋亡。 Objective To investigate the effect of panax notoginseng saponis(PNS) on proliferation and apoptosis of human acute monocytic leukemia cell line U937 and the expression of caspase-3 in it. Methods U937 cells were treated with PNS at different concentrations. Cell proliferation was analyzed by MTT assay. Cell cycle and apoptosis was assessed by flow cytometry, and the expression of Caspase-3 protein was detected by Western blot. Results The inhibition of cell proliferation in U937 cells exposed to PNS was in a dose and time dependent manner. At 48 hours after exposure to 100,200,400,800 mg/L PNS,the apoptotic rates of U937 cells were (5.93±0.15)%,(7.43±0.70)%,(12.3±0.38)% and (20.4±0.91)%,respectively,which were markedly higher than that of control group(1.73±1.50%,P〈0.01). The exposure to PNS induced the G1 phase arrest of U937 cells and up-regu-lated the expression of Caspase-3 protein in U937 cells. Conclusion PNS can inhibit the proliferation of U937 cells and induce apoptosis by up-regulating the expression of Caspase-3.
