摘要
目的:研究绞股蓝总皂苷对光老化人皮肤角质形成细胞分泌炎症因子IL-1β、IL-6和TNF-α表达的影响。方法:选取10只大鼠,随机分为2组,每组5只,分别设为空白血清组、绞股蓝总皂苷含药血清组,分别用以制备空白大鼠血清和绞股蓝含药血清。应用UVB人工照射方法制备光老化人皮肤角质形成细胞模型,用含绞股蓝总皂苷的大鼠血清进行干预,12h后采用ELISA方法检测各组细胞培养上清液中IL-1β、IL-6和TNF-α的水平。结果:与空白对照组比较,UV模型组细胞IL-1β、IL-6和TNF-α表达显著上调(P<0.01);与UV模型组比较,绞股蓝总皂苷含药血清组细胞中IL-1β、IL-6和TNF-α表达均显著下调(P<0.01);与空白血清组比较,绞股蓝总皂苷含药血清组细胞中IL-1β、IL-6和TNF-α表达均显著下调(P<0.01)。结论:绞股蓝总皂苷可抑制UV辐射人皮肤角质细胞分泌炎症因子,抑制皮肤光老化。
Objective: To research the effect of gypenosides-containing serum on secretion of inflammatory factors IL-1β,IL-6 and TNF-α of photoaging skin HaCaT cells. Methods: 10 rats were randomly divided into 2 groups with 5 rats in each group:blank serum group,gypenosides-containing serum group. After UVB irradiation,the artificial light aging of HaCaT cell models was interfered with rat gypenosides-containing serum group. 12 h later,the expression levels of cell secretion of IL-1β,IL-6and TNF-α were determined with ELISA. Results: Compared with the blank control group,IL-1β,IL-6 and TNF-α expressions of UVB model group were significantly higher( P 〈 0. 01). Compared with UVB model group,IL-1β,IL-6 and TNF-αexpressions in cells of gypenosides-containing serum group were significantly lower( P 〈 0. 01). Compared with blank serum group,IL-1β,IL-6 and TNF-α expressions in cells of gypenosides-containing serum group were significantly lower( P 〈 0.01). Conclusion: Gypenosides can protect skin photo aging by inhibiting the UVB radiation of human skin HaCaT cells to secrete inflammatory factors.
