摘要
以百脉根的下胚轴为外植体材料,探究不同激素种类和不同浓度对百脉根愈伤组织的诱导、分化增殖及生根的影响.结果表明:诱导愈伤组织的最佳培养基为:MS+2,4—D 2.0 mg/L+KT 2.0 mg/L+3%蔗糖+0.7%琼脂;诱导愈伤组织分化最佳培养基为:MS+6-BA 1.0 mg/L+NAA 0.5 mg/L+3%蔗糖+0.7%琼脂;诱导不定芽生根的最佳生根培养基为:MS+NAA 0.01 mg/L+2%蔗糖+0.7%琼脂,生根率达100%;实验建立了一个高频的百脉根再生体系,为百脉根基因工程操作奠定了基础.
The hypocotyl of the Lotus corniculatus was taken as the explants for researching the effects of different hormone compositions on callus induction, plantlet regeneration and plantlet roots. The results showed that the medium of MS + 2. 4 - D 2.0 mg/L + KT 2.0 mg/L + 3 % sucrose + 0. 7% agar could efficiently induce callus tissue; and the medium of MS + 6 - BA 1.0mg/L + NAA 0. 5mg/L + 3% sucrose + 0. 7% agar could efficiently induce bud of callus; medium of MS + NAA 0. 1 mg/L + 2% sucrose + 0. 7% agar induced the regeneration of roots quickly ,and the rooting rate reached 100%. This research built the best regeneration system that had laid a sound foundation for transgenic opecation of Lotus corniculatus.
