摘要
目的观察土贝母皂苷对转染绿色荧光蛋白(GFP)基因的人乳腺癌细胞MDA-MB-231、MCF-7的体外抗肿瘤活性。方法采用倒置光学显微镜观察土贝母皂苷作用下转染GFP基因的乳腺癌细胞MDAMB-231、MCF-7形态学变化,MTT法检测土贝母皂苷对两种乳腺癌细胞的生长抑制作用。结果土贝母皂苷作用24h后,两种人乳腺癌细胞均表现出典型凋亡形态改变;MTT结果显示土贝母皂苷对两种乳腺癌细胞的生长具有强抑制作用,MDA-MB-231(GFP)、MCF-7(GFP)经土贝母皂苷作用48h后的IC50分别为(17.03±3.22)、(27.96±0.34)μg/ml。结论土贝母皂苷对两种人乳腺癌细胞均有较强的抑制作用。
Objective To research the antitumor activity in vitro of tubeimoside on human breast cancer cell lines MDA-MB-231 and MCF-7 transfeeted with green fluorescent protein (GFP). Methods The morphological changes of MDA-MB-231 (GFP) and MCF-7 (GFP) under the action of tubeimoside were observed with inverted mi- croscope. The inhibitory effect of tubeimoside on the growth of MDA-MB-231 (GFP) and MCF-7 (GFP) were meas- ured by MTT assay. Results A typical apoptosis of MDA-MB-231 (GFP) and MCF-7 (GFP) were shown 24 hours later. MTY assay showed that tubeimoside had a strong inhibitory effect on the growth of MDA-MB-231 (GFP) and MCF-7 (GFP). The inhibitory concentration 50 (IC50) of MDA-MB-231 (GFP) and MCF-7 (GFP) was ( 17.03 ± 3.22) μg/ml and (27.96±0. 34 ) μg/ml respectively 48 hours later. Conclusion Tubeimoside has a strong inhibitory effect on the growth of MDA-MB-231 (GFP) and MCF-7 (GFP)
