萎胃颗粒对慢性萎缩性胃炎大鼠胃黏膜Bcl-2和PCNA表达的影响

Effects of Weiwei Granules on the Expressions of Bcl-2 and PCNA in Mucosa of Rats With Chronic Atrophic Gastritis

目的探讨萎胃颗粒治疗慢性萎缩性胃炎(CAG)的可能作用机制。方法将50只SD大鼠随机分为正常组、模型组和萎胃颗粒低、中、高剂量组,每组10只。除正常组外,其余各组大鼠复制CAG模型,造模后正常组和模型组给予10ml/kg自来水灌胃,萎胃颗粒低、中、高剂量组分别给予萎胃颗粒1.8、3.6、7.2g/d灌胃,共连续12周。采用免疫组织化学方法检验各组大鼠胃黏膜增殖细胞核抗原(PCNA)和抑凋亡基因(Bcl-2)表达。结果与正常组比较,模型组大鼠胃黏膜Bcl-2、PCNA表达显著增多(P<0.01);与模型组比较,萎胃颗粒各治疗组Bcl-2均有明显下降(P<0.01);与萎胃颗粒低剂量组比较,中剂量组Bcl-2下降(P<0.01);与萎胃颗粒中剂量组比较,高剂量组Bcl-2下降(P<0.01)。与模型组比较,萎胃颗粒各治疗组的PCNA阳性表达率均有明显下降(P<0.01);与萎胃颗粒低剂量组比较,中剂量组阳性表达率下降(P<0.01);与萎胃颗粒中剂量组比较,高剂量组阳性表达率下降(P<0.01)。结论萎胃颗粒可能通过抑制CAG大鼠胃黏膜Bcl-2和PCNA的表达,调节细胞的增殖和凋亡,从而改善CAG胃黏膜病变。

Objective To research the effects and mechanism of Weiwei Granules on chronic atrophic gastritis （CAG）. Methods Totally 50 SD rats were randomized into the normal group, model group and Weiwei Granules low-dose （1.8 g/d） , middle-dose （3.6 g/d） and high-dose （7.2 g/d） groups, with 10 in each. The CAG models were established in all groups except the normal group with comprehensive method. The intragastric administration las- ted for 12 weeks. The expressions of Bcl-2 and proliferating cell nuclear antigen （PCNA） in mucosa of CAG rats were observed with immunohistochemical method. Results Comparing with the model group, the OD values of Bcl-2 in Weiwei Granules groups were significantly decreased （P 〈 0.01 ）. Comparing with the Weiwei Granules low-dose group, the OD value was significantly decreased in the middle-dose group （P 〈 0. 01 ）. Comparing with the Weiwei Granules middle-dose group, the OD value was significantly decreased in the high-dose group （P 〈 0. 01 ）. Comparing with the model group, the positive expression rate of PCNA in Weiwei Granules groups were significantly decreased （P 〈 0. 01 ）. Comparing with the Weiwei Granules low-dose group, the positive expression rate of PCNA was significant- ly decreased in the middle-dose group （P 〈0. 01 ）. Comparing with the Weiwei Granules middle-dose group, the posi- tive expression rate of PCNA was significantly decreased in the high-dose group （ P 〈 0. 01 ）. Conclusion Weiwei Granules can relieve the pathological state of gastric mucosal lesions and avoid stomach cancer possibly by inhibiting the expressions of Bcl-2 and PCNA in mucosa of CAG rats and regulating the proliferation and apoptosis of cells.
