直接皂化法制备叶绿素铁钠盐

摘要 以蚕沙为原料,采用直接皂化法制备叶绿素铁钠盐,通过单因素实验和正交试验设计研究了体系pH值、超声时间、超声温度、置铁量、置铁时间、置铁温度对叶绿素铁钠盐产率的影响,其最优工艺条件为:10 g干蚕沙粉先在p H≈12的体系中,80℃下超声120 m in ;再于pH≈2~3的体系中,65℃下酸化30 min ,加入6 mL10% FeSO4溶液,置铁时间150 min ,最后用10% NaOH-CH3CH2OH调节溶液使叶绿素铁酸成盐析出。通过对照实验,直接皂化法叶绿素铁钠盐的得率大于传统制备法叶绿素铁钠盐的得率,而且直接皂化法比传统制备法在制备工艺上还少一道工序,因此直接皂化法是可行的。 Sodium iron chlorophyllin was prepared through the reaction of direct saponifica-tion ,using silkworm faeces as raw material .Effects of factors ,such as system pH ,ultrasonic time ,ultrasonic temperature ,dosage of iron ,time on the replacement of sodium by iron ,tem-perature on the replacement of sodium by iron on the yield of sodium iron chlorophyllin were investigated by single factor test and orthogonal experimental design .The optimal process conditions were identified as :Firstly ultrasound 10 g dried silkworm powder for 120 min at 80 ℃ in the system with pH≈12 ;Secondly acidification for 30 min at 65 ℃ in the system with pH≈2~3 ,and after replace sodium with 6 mL 10% FeSO4 solution for 150 min ;Final-ly ,the sodium iron chlorophyllin was obtained by neutralizing at pH≈12 with 10% NaOH-ethanol .Through contrast test ,the yield of sodium iron chlorophyllin by direct saponification was superior to the traditional one ,and the preparation procedure of direct sapinification was less than the traditional one ,so direct saponification was feasible .
