广西眼镜王蛇毒酸性磷脂酶A_2的肌毒性研究 被引量:6

摘要 目的 研究广西眼镜王蛇毒一种酸性磷脂酶 A2 (命名为 APLA2 -1 )的肌毒性。 方法 大鼠注射 2 .6 mg/ kg的 APLA2 -1后 ,分别于 6 h、2 4 h取心肌和骨骼肌通过光镜和电镜观察它们的病理改变 ;或于0 h、 3h、 6 h、 1 2 h、 2 4 h取尾静脉血测定血清肌酶的动态变化。 结果 光镜检查显示 ,大鼠注射 APLA2 -1 2 4 h后 ,骨骼肌发生变性和轻度坏死 ,心肌无明显改变 ;电镜结果显示 ,骨骼肌在 6 h、 2 4 h,心肌在 2 4 h,细胞超微结构均受到不同程度的破坏 ;血清肌酶检测显示 ,注射 APLA2 -1后 ,肌酶升高显著 ,1 2 h达到最大值。 结论 广西眼镜王蛇毒酸性磷脂酶 A2 具有肌毒性 ,骨骼肌比心肌对其更为敏感 ;与别的肌毒性PLA2 相比 。 Objective This paper reports the myotoxic activity of an acidic phospholipase A 2 from Guangxi Ophiophagus hannah venom(signed as APLA 2 1) Methods Mice were injected intravenously with dose of 2 6 mg/kg of APLA 2 1 Then their tissue samples were taken to examine pathological changes at 6 or 24 h,or their serum myokinase were assayed at 0h,3h,6h,12h or 24h respectively Result Light microscopic examination showed degeneration and low grade necrosis in skeletal muscle at 24 h but failed to showed significant changes in cardiac muscle Electron microscopic study revealed the ultrastructure of skeletal muscle and cardiac muscle were all destroyed in some extent The skeletal muscle were more severe After injected APLA 2 1,the mice' serum myokinase rose markedly and reached the highest value in 12h Conclusion APLA 2 1 has myotoxic activity The skeletal muscle were more sensitive to APLA 2 1 Compared with some myotoxic PLA 2,the toxicity of APLA 2 1 was relative slow and gentle [
