摘要
目的探讨纳米硫化镉(nano-CdS)对人肾小球系膜细胞(HMC)的氧化损伤及其机制。方法将对数生长期的HMC细胞分为对照组(0μg/ml)和nano-CdS组(5~80μg/ml),用MTT实验观察nano-CdS对细胞存活率的影响,采用荧光探针技术检测细胞内的活性氧(ROS),采用蛋白质印迹法观察NF-κB蛋白的表达。结果与对照组比较,nano-CdS组细胞存活率降低,ROS含量升高,NF-κB的蛋白表达增加,差异均有统计学意义(P〈0.05)。结论 nano-CdS可导致HMC细胞氧化损伤。
Objective To investigate the cytotoxicity of nano-cadmium sulfide(nano-CdS) on human glomerular mesangial cells(HMC). Methods After HMC cells were exposed to different concentrations of nano-CdS(5-80 μg/ml) for 24 h, cell viability were observed by MTT assay. The content of ROS and level of NF-κB protein in cells were determined by fluorescence probe and Western blotting individually. Results Compared with the control group, the viability of HMC cells was decreased with increasing concentrations of nano-CdS(P〈0.05). After exposed to nano-CdS for 24 h, as the concentration increased, the content of ROS and NF-κB protein was enhanced(P〈0.05). Conclusion Cell toxicity of nano-CdS may be due to the mechanism involving antioxidant system damage.
