黄芪多糖对拟缺血损伤内皮祖细胞保护作用的实验研究 被引量:2

摘要 目的通过动物实验,评价黄芪多糖对拟缺血内皮祖细胞的保护作用。方法采用凝血酶损伤内皮祖细胞复制大鼠部分缺血损伤模型,不同浓度黄芪多糖预干预内皮祖细胞24h、凝血酶损伤8h后,用四甲基偶氮唑盐染色法检测细胞活性,蛋白印迹法和聚合酶链反应检测内皮祖细胞在蛋白及基因水平表达的变化。结果凝血酶损伤后,与正常对照组相比,凝血酶损伤组细胞活性明显下降,Ang-1及其受体表达下降(P〈0.05);与凝血酶损伤组比较,黄芪多糖各组细胞活性上升, Ang-1及其受体表达上调(P〈0.01);黄芪多糖各剂量组间呈剂量依赖。结论凝血酶可诱导内皮祖细胞损伤,黄芪多糖可抑制凝血酶所致体外培养的EPCs的损伤,对内皮祖细胞有保护作用,其机制与增加Ang-1及其受体表达有关,为缺血性疾病提供了新的治疗思路。
