脯氨酸对杉木愈伤组织超低温冷冻保存的影响

摘要 为探索脯氨酸在杉木胚性愈伤组织超低温冷冻保存过程中的作用,本试验以继代14 d的杉木愈伤组织为材料,在预处理阶段设置不同浓度的脯氨酸,采用TTC法测定相应的细胞存活率。结果表明:预处理阶段,脯氨酸浓度为0.5 g·L-1时,细胞存活率相对最高。说明了在预处理阶段添加适宜浓度的脯氨酸可提高超低温冷冻保存的存活率。 To explore the role of prolin in the process of cryopreservation of embryogenic callus of Cunninghamia lanceolata,the cell survival of 14 days callus of Cunninghamia lanceolata of subculture was measured by the TTC method with different concentrations of proline in the pretreatment stage.The result showed that:in the pretreatment stage,when the concentration of proline was 0.5 g·L-1,the cell survival was relatively the highest.It suggested that adding suitable concentration of proline in the pretreatment stage can improve the survival of cryopreservation.
