摘要
目的 测定68例小细胞肺癌中UGT1A1* 28基因多态性的频率与分布情况.方法 收集中南大学湘雅医院68例中国人小细胞肺癌患者和33名中国正常人外周血标本,抽提基因组DNA,PCR法扩增目的基因片段,荧光标记测序法测定UGT1A1基因启动子区TATA盒胸腺嘧啶-腺嘌呤(TA)序列重复次数,统计中国小细胞肺癌和正常人中UGT1A1* 28基因多态性分布情况.结果 68例小细胞肺癌患者中51例(75.00%)为(TA) 6/(TA)6型,17例(25.00%)为(TA) 6/(TA)7型,0例为(TA)7/(TA)7型;33名正常人中25名(75.76%)为(TA) 6/(TA)6型,6名(18.18%)为(TA)6/(TA)7型,2名(6.06%)为(TA)7/(TA)7型.结论 UGT1A1* 28基因多态性在中国小细胞肺癌与正常人群中的分布有统计学差异.中国人UGT1A1* 28基因多态性发生频率与欧美人群有较大区别.
Objective To determine the frequency and distribution of UGT1A1 * 28 polymorphism in 68 patients with small cell lung cancer in China.Methods Peripheral blood samples were collected from 68 cases of small cell lung cancer and 33 normal persons.Genomic DNA was extracted from the patients leukocytes in the peripheral blood.Then the target gene fragment was amplified by PCR and the genotypes were determined by analyzing the sequence of TATA boxes in the UGT1A1 gene by fluorescently labeled sequencing method.The frequency and distribution of UGT1A1 * 28 polymorphism in patients with small cell lung cancer and normal persons in China were analyzed.Results In 68 cases of small cell lung cancer:51 patients (75.00%) with (TA)6/(TA)6 genotype,17 patients (25.00%) with (TA)6/(TA)7 genotype,nopatients with (TA)7/(TA)7 genotype;in 33 normal persons:25 persons (75.76%) with (TA)6/(TA)6 genotype,six persons (18.18%) with (TA) 6/(TA) 7 genotype,two persons (6.06%) with (TA)7/(TA)7 genotype.Conclusions There is significant difference on the distribution of UGT1A1 * 28 polymorphism in patients with small cell lung cancer and normal population in China.The incidence of UGT1A1 * 28 polymorphism in China is significantly different from European and American.
