Effect of down- regulation of AEG-1 expression by siRNA on proliferation and apoptosis of breast cancer cells

摘要 目的:探讨si RNA下调AEG-1表达对乳腺癌细胞增殖和凋亡的影响。方法:用设计合成的AEG-1 siRNA转染人乳腺癌MCF-7细胞株,采用荧光定量RT-PCR技术观察AEG-1 siRNA对MCF-7细胞AEG-1 mRNA表达的影响;MTT法检测AEG-1 siRNA对MCF-7细胞增殖的抑制作用;流式细胞术检测AEG-1 siRNA对MCF-7细胞凋亡和细胞周期的影响。结果:AEG-1 siRNA转染人乳腺癌MCF-7细胞后能显著下调AEG-1基因的表达,与对照组比较差异有统计学意义(P<0.01);AEG-1基因表达的下调可以显著抑制MCF-7细胞的增殖,与对照组比较差异有统计学意义(P<0.01),促进MCF-7细胞凋亡,与对照组比较差异有统计学意义(P<0.01),并将细胞周期阻滞在G0/G1期,与对照组比较差异有统计学意义(P<0.01)。结论:AEG-1 si RNA可下调乳腺癌MCF-7细胞AEG-1基因的表达,抑制其增殖,并促进其凋亡。 Objective: To explore the effect of down - regulation of astrocyte elevated gene - 1 ( AEG - 1 ) expression by siRNA on proliferation and apoptosis of breast cancer cells. Methods : AEG - 1 siRNA was used to transfect human breast cancer MCF - 7 cells, fluo- rescent quantitative RT - PCR was used to observe the effect of AEG - 1 siRNA on AEG - 1 mRNA expression in human breast cancer MCF - 7 cells ; MTY method was used to detect the inhibiting effect of AEG - 1 siRNA on proliferation of human breast cancer MCF - 7 cells ; flow cytometry was used to detect the effect of AEG - 1 siRNA on apoptosis and cell cycle of human breast cancer MCF - 7 cells. Re- suits : AEG - 1 siRNA down - regulated the expression of AEG - 1 mRNA significantly in human breast cancer MCF - 7 cells, compared with control group, there was statistically significant difference (P 〈0.01 ) ; down- regulation of AEG- 1 mRNA inhibited the proliferation of human breast cancer MCF- 7 cells significantly, compared with control group, there was statistically significant difference (P 〈 0. 01 ) ; down - regulation of AEG - 1 mRNA promoted the apoptosis of human breast cancer MCF - 7 cells significantly, compared with control group, there was statistically significant difference ( P 〈 0. 01 ) ; down - regulation of AEG - 1 mRNA arrested cell cycle at G0/G1 phase, compared with control group, there was statistically significant difference (P 〈 0. 01 ) . Conclusion: AEG - 1 siRNA can down - regulate the expression of AEG - 1 mRNA in human breast cancer MCF - 7 cells, inhibit cell proliferation and promote cell apoptosis.
