摘要 采用免疫印迹和间接免疫荧光方法,对荷斯坦奶牛不同发育时期乳腺组织IGF-I及其受体IGF-IR的表达与定位进行研究,结果表明,IGF-I(Insulin-like growth factor-I)及IGF-IR(Insulin-like growth factor-I receptor)均定位于乳腺导管腔上皮细胞、腺泡上皮细胞以及基质;IGF-I蛋白表达量在围产前期降到最低,在泌乳中期达到最高;IGF-IR蛋白表达量在妊娠6月急速下降,随后在围产前期上升至最大值。结论:IGF-I与IGF-IR蛋白在奶牛乳腺中定位的一致性及其特异性表达规律,预示配体与受体相结合并参与了奶牛乳腺发育及泌乳的调控。 To investigate the expressions and localizations of IGF-I and IGF-IR at different developmental stages in Holstein dairy cow mammary gland, the immunofluorescence assay and Western blotting method were used. Results showed that IGF-I and IGF-IR localized in the mammary gland luminal epithelial cells, acinar cells and matrix. The expression of IGF-I protein was lowest at peripartum, and was highest in the middle of lactation. The expression of IGF-IR protein decreased at P6m, and then increased to the highest at peripartum. Conclusion: The consistent localizations of IGF-I and IGF-IR proteins and their specific expressions indicated they played a role in the regulation of development and lactation of dairy cow mammary gland.
