摘要
为增加黑糯种质材料,以甲基磺酸乙酯(EMS)诱变处理的贵州地方稻种平塘黑糯得到的半矮秆突变体,并对突变体相关性状进行观察测定。结果表明:1)筛选的黑糯稻半矮秆突变体bgsd-2株高为65.20cm、剑叶长22.24cm、总穗粒数166粒、有效粒数147粒,与平塘黑糯差异极显著;千粒重25.76g,与平塘黑糯差异显著;其余性状两者无显著性差异。2)突变体bgsd-2对外源GA敏感,微管免疫荧光定位检测发现外源GA3能恢复突变体bgsd-2根尖倾斜排列的微管至平塘黑糯排列方式。3)突变体bgsd-2与9311、平塘黑糯杂交的F1代均表现为高秆,显示bgsd-2为隐性突变体。
In order to obtain new rice dwarf mutants,a semi-dwarf mutant,bgsd-2 was identified from mutant progeny of the Guizhou local black glutinous cultivar rice by ethyl methanesulfonate (EMS ) treatment.In this study,the morphology and cytology of bgsd-2 were investigated and analyzed.Results:1)bgsd-2 mutant was a semi-dwarf grains,the plant height was 65.20 cm,the Flag leaf length was 22.24 cm,the seeds number per panicle was 166,the fertile seeds number per panicle was 147,their differences reached the very significant level with the Guizhou local black glutinous cultivar rice,while 1000-grain weight reached the significant level.There was no significant difference among the other characters.2 ) The bgsd-2 mutant was sensitive to response of exogenous GA3 . Immunolocalization of cortical microtubule (CMT ) assays found that the oblique CMTs abnormal organizations in bgsd-2 could be restored after exogenous application of GA3 .The two mutants were crossed with 9311 and wild-type,all F1 progeny showed a height phenotype.These showed that bgsd-2 mutant was the cryptic mutant.
