摘要
目的对聚乙二醇-聚谷氨酸苄酯两亲嵌段共聚物的细胞毒性进行考察。方法采用四噻唑蓝比色法、聚乙二醇-聚谷氨酸苄酯对体外HeLa细胞的毒性进行研究,并与增溶剂聚氧乙烯蓖麻油进行比较。结果聚乙二醇-聚谷氨酸苄酯对HeLa细胞的毒性远低于聚氧乙烯蓖麻油,仅在400μg·mL-1浓度对HeLa细胞的细胞相对增殖百分率为62.8%,表现为轻度细胞毒性。结论与增溶剂聚氧乙烯蓖麻油相比,聚乙二醇-聚谷氨酸苄酯嵌段共聚物细胞毒性极低,在医学研究上有较为广阔临床应用前景。
Objectives To study the cell cytotoxicity of the Polyethylene glycol-polybenzyl-L-glutamate, ( PEG-PBLG ). Methods The cell cytotoxicity of PEG-PBI.G on HeLa cells was investigated by comparing with Gremophor El. by MTT way. Results The results showed that the cell cytotoxicity of PEG-PBLG had much less than Cremophor El.. Only at 400 μg·mL^-1 high concentration, PEG- PBLG was low cell cytotoxicity. Conclusion The PEG-PBLG had less cytotoxicity than Crcmophor EL, so it might be a novel block copolymers preparation for clinical therapy.
