摘要 目的探讨O6-甲基鸟嘌呤-DNA甲基转移酶(MGMT),信号转录和转导激活因子3(STAT-3)在中枢神经系统星型细胞瘤组织中的表达情况及其临床意义。方法收集120例手术切除的原发中枢神经系统星型细胞瘤组织与25例正常人脑组织,应用免疫组化方法检测全部标本中MGMT和STAT-3的表达情况,并进行相关性分析。结果 MGMT,STAT-3经免疫组织化学染色后在正常组织中表达阴性,在肿瘤标本中呈阳性表达,表达率随星型细胞瘤病理级别升高,呈上升趋势。低级别组与高级别组差异显著。相邻组间无显著差异。相关性分析显示二者的表达存在正相关。结论结果提示MGMT和STAT-3在人中枢神经系统星型细胞肿瘤中的表达与肿瘤的恶性进展具有相关性。STAT-3可能参与调控MGMT的表达。 Objective This paper aims to investigate the expressions and clinical significance of O6-mehtylguanine DNA methyltransferase (MGMT)and signal transduction andactivators of transcription-3 (STAT-3) in astrocytoma of central nervous system.Methods A total of 120 cases of human astrocytoma tissues and 25 cases of normal brain tissues were collected.Expressions of MGMT and STAT-3 and the correlation between them were studied by immunohistochemistry.Results Negative expression of MGMT and STAT-3 was found in normal brain tissues by immunohistochemistry while positive expression was found in the astrocytoma specimens,and the expressions were increased with the increasing of malignancy.There was significant difference in the expressions of MGMT and STAT-3 between low grade groups and high grade groups (P 〈0.05).No significant difference was found in the adjacent groups.The expression of STAT3 was positively related to that of MGMT.Conclusion The results indicate that the expressions of MGMT and STAT-3 in human astrocytomas are related to the malignant progression of tumors.STAT-3 may play a certain role in the regulation of MGMT expression.
