摘要
为了建立奶牛外周血单个核细胞增殖反应的MTT检测方法,对影响增殖反应的刺激原及浓度、细胞浓度、培养时间等条件进行了探索性研究.结果表明,MTT法检测的最佳条件是:ConA浓度为2.5 μg/mL,细胞浓度为1×10^6 cells/mL,培养时间为48 h;或者浓度为30 μg/mL,细胞浓度为1×10^6 cells/mL,培养时间为48 h.
In order to establish a MTI" assay detecting the proliferation response of peripheral blood mononuclear cell (PBMC), three major factors including the stimulus and its concentration, cell concentration and culture time were studied. The results showed that the best condition of proliferation response was 2.5 μg/mL of ConA concentration, 1 × 10^6 cells/mL of cell concentration, and 48 h of culture time, or 30 μg/mL of LPS concentration, 1 × 10 ^6 cells/mL of cell concentration and 48 h of culture time.
