PD-118057药物拯救负显性机制E637K-hERG通道的研究

摘要 目的探讨PD-118057对于纠正负显性机制E637K-hERG蛋白转运障碍的作用。方法全细胞膜片钳技术检测PD-118057干扰各种细胞模型前后hERG编码的快速激活延迟整流性钾电流(IKr)变化;采用Western blot分析PD-118057负显性机制E637K-hERG蛋白转运障碍。结果 PD-118057干扰WT-hERG后,激活电流和尾电流的电流幅度都较干扰前明显增大,但不改变通道电流的特性,只加速了通道电流的稳态失活速度。而PD-118057干扰WT/E637K-hERG前后电流幅度和通道特性都没有明显变化。Western blot结果与膜片钳结果相符合。结论 PD-118057不能纠正负显性机制E637K-hERG通道功能。
