成骨相关基因在强直性脊柱炎与正常人成纤维细胞的表达差异 被引量:2

摘要 目的:探究强直性脊柱炎(Ankylosing Spondylitis,AS)患者与正常人脊上韧带成纤维细胞的原代爬出时间、生长曲线及两组细胞之间的成骨相关性指标的差异。方法:取AS和正常人棘上韧带,离体原代培养成纤维细胞。记录组织爬出成纤维细胞时间和四甲基偶氮唑盐(MTT)检测法检测两组细胞生长曲线。原代细胞爬出后传代培养,取对数期细胞RNA并逆转录CDNA,实时荧光定量PCR法检测两组细胞之间成骨相关基因表达的差异。结果:两组细胞的爬出时间有明显差异(P<0.05),生长曲线大致成"S"型,两组细胞生长曲线未见明显差异。Ⅰ型胶原酶(COL1A1)在两组之间表达无差异(P>0.05),骨钙素(OC)基因起始表达量过低,可记为不表达。结论:AS组与正常人组成纤维细胞成骨性相关基因原始表达量和生长曲线均无明显差异。
