摘要
目的:建立偏头痛大鼠皮层扩布性抑制(cortical spreading depression,CSD)模型,研究CSD对大鼠血浆降钙素基因肽(calcitonin gene-related peptide,CGRP)和P物质(substance P,SP)的影响。方法:大鼠随机分为CSD组(n=10)、对照组(n=10),分别用氯化钾溶液及生理盐水浸润滤纸片刺激枕叶皮层,在额叶皮层记录CSD波,利用放射免疫技术测定大鼠血浆中的CGRP、SP水平,进行成组t检验比较。结果:CSD组大鼠记录到CSD波,对照组大鼠未记录到CSD波;CSD组大鼠血浆CGRP、SP水平高于对照组(P<0.05)。结论:利用氯化钾溶液浸润滤纸片刺激大鼠枕叶皮层成功建立了CSD模型,CSD使血浆CGRP、SP水平增加,说明CSD可能通过一定的机制激活了三叉神经血管反射,进而导致偏头痛的发生。
Objective:A rat cortical spreading depression (CSD) model of migraine was established to explore the effect of CSD on calcitonin gene-related peptide (CGRP) and substance P (SP) levels in serum.Methods:Twenty adult rats were randomly and evenly divided into two goups,CSD Group and Normal Control Group.The CSD waves were evoked by application of potassium chloride on brain surface with filter paper.For control group,normal saline,instead of potassium chloride,was used for stimulation on brain surface with filter paper.The levels of CGRP and SP in plasma were measured by radioimmunity assay.Statistical analysis was performed using two-sample t test.Results:CSD waves were induced in rats of CSD Group.There was no CSD wave in rats of Normal Control Group.The levels of both CGRP and SP in CSD Group were higher than the Control Group (P 〈 0.05).Conclusion:The rat CSD model of migraine was successfully established.CSD increases the serum level of CGRP and SP.It is implicated that CSD might activate peripheral trigeminovascular reflex by unknown mechanisms,resulting in the migraine attacks.
