硫酸右旋糖苷对人胃癌细胞腹腔种植转移和VEGF表达的影响 被引量:3

Effects of dextran sulfate on expression of VEGF and human gastric cancer cell metastasis in abdominal cavity

摘要 目的通过观察人胃癌细胞在裸鼠大网膜上的种植转移情况和测定血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达水平,探讨硫酸右旋糖苷(dextran sulfate,DS)对人胃癌细胞腹腔种植转移和VEGF表达的影响。方法采用细胞培养,建立动物模型,将108只裸鼠随机分为对照组与实验组两组,在注射胃癌细胞的同时分别注射生理盐水与DS。剖腹观察腹腔种植的癌结节数量,应用免疫组化和RT-PCR法检测VEGF蛋白及mRNA的表达。结果实验组癌结节数明显少于对照组(P<0.001),免疫组化和RT-PCR检测结果均显示实验组VEGF蛋白及mRNA的表达量明显小于对照组。结论DS可以抑制VEGF的表达并有可能通过抑制其表达降低胃癌的腹腔种植转移。 Purpose To observe the implanting metastasis of human gastric cancer cells into nude omentum, to determine the level of vascular endothelial growth factor (VEGF) expression, and to explore the effects of dextran sulfate (DS) on the implanting metastasis of human gastric cancer cells into nude omentum and the expression of VEGF. Methods Using cell culture , cultured cells, an animal model of nude mice was established. 108 nude mice were randomly divided into control and experimental groups, both of which were injected with normal saline and dextran sulfate with the tumor cells into abdominal cavity in the same time. Peritoneal cancer nodule number was observed by caesarean section. The expression levels of VEGF mRNA and protein were detected by immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR). Results In the experimental group, the number of implanted tumor nodules was significantly less than those of the control group (P 〈0. 001 ). Immunohistochemistry and RT-PCR showed that the expression of VEGF protein and mRNA in the experimental group was significantly less than that of the control group. Conclusion DS can inhibit the expression of VEGF and may reduce peritoneal metastasis of gastric cancer by inhibiting the expression of VEGF.
