摘要 目的建立大鼠C6胶质瘤模型,探索胶质瘤模型建立的实验步骤、方法及关键点。材料与方法大鼠C6胶质瘤细胞体外培养、传代至对数生长期,离心、计数,调节细胞浓度为1×10^5/μl。应用大鼠立体定位仪,以右侧尾状核头为靶区,注入Wistar大鼠脑内,3d后行MRI扫描观察脑内成瘤情况。本实验共接种8批次50只Wistar大鼠。结果第1~5次共接种24只Wistar大鼠,接种后第1、2天意外死亡2只,其余22只大鼠MRI扫描可见肿瘤生长,成瘤率高。第6、7次以同样方法及条件接种20只大鼠,MRI扫描证实仅有5只大鼠成瘤。第8次更换肿瘤细胞后接种6只大鼠,均成瘤。结论肿瘤是否成瘤与细胞选择密切相关,要选择新近诱导出的大鼠胶质瘤细胞,以保证细胞的抗原性,是提高成瘤率的关键。 Purpose To explore the procedure, method and key factors in establishing rat glioma model. Materials and Methods C6 glioma cells were cultured in vitro to exponential phase. The cells were centrifuged and adjusted to the concentration of 1 × 10^5/μl, and then implanted into the right caudate head in Wistar rats using stereotaxic positioner. MRI was performed in three days to evaluate tumor formation. Fifty rats were inoculated in eights batches. Results In the first five batches, 24 Wistar rats were injected. Two rats died on day 1 and 2. In the remaining 22 rats, tumors were demonstrated on MRI indicating high tumor formation rate. Twenty rats were injected in batch 6 and 7 with tumor formation in only 5 rats on MRI. In batch 8, new C6 tumor cells were used with tumor formation in all 6 rats. Conclusion Successful tumor growth is closely related to cell selection. Recently inducted C6 rat glioma cells are crucial to maintain antigenicity and tumor formation.
