摘要
目的:观察腰痹通胶囊对兔退变腰椎间盘Ⅱ型胶原含量和TNF-α、IL-1βmRNA表达的影响。方法:采用纤维环针刺法复制兔腰椎间盘退变动物模型。造模成功后,将48只新西兰大白兔随机分为腰痹通、腰痛宁、美洛昔康、模型对照组4组,给药6周,处死后取退变的腰椎间盘,采用免疫组化染色法测定Ⅱ型胶原含量,RT-PCR法检测TNF-α、IL-1βmRNA的表达情况。结果:腰痹通、腰痛宁组Ⅱ型胶原的含量较美洛昔康和模型对照组均有明显提高,相比差异具有统计学意义(F=9.41,P=0.008<0.01)。与模型组比较,腰痹通、腰痛宁、美洛昔康组TNF-α、IL-1βmRNA的表达均下调(F=10.32,P=0.006<0.01);腰痹通、腰痛宁、美洛昔康组TNF-α、IL-1βmRNA的表达比较差异没有统计学意义(F=3.26,P=0.086>0.05)。结论:腰痹通胶囊提高了椎间盘中Ⅱ型胶原的含量,并下调了TNF-α、IL-1βmRNA的表达,减少了炎症介质的合成,这可能是腰痹通胶囊减缓椎间盘退变,治疗椎间盘退行性疾病的机制之一。
Objective:To observe the effects of Yaobitong Capsule on the content of type II collagen and expression of IL-1β and TNF-α mRNA in rabbit degenerated lumbar intervertebral disc. Methods: After successful reproduction of disc degeneration animal model by an annulus needle puncture, 48 New Zealand rabbits were randomly and equally divided into 4 groups, i.e. Yaobitong Capsule group, Yaotongning Capsule group, Meloxicam Capsule group and model control group. Then each group was given the corresponding medicines for 6 weeks. Immuno-histochemical staining method was used to detect the content of type II collagen in the intervertebral disc. The expression of IL-1β and TNF-α mRNA was detected by RT-PCR. Results:Compared with Meloxicam Capsule and model control groups, the content of type II collagen of Yaobitong and Yaotongning Capsule groups were higher (F=9.41, P=0. 008〈0.01). The expression of IL-1β and TNF-α mRNA of the model control group were higher compared with the other three groups respectively (F= 10.32, P= 0. 006%0. 01) . There was no significant difference in the expression of IL-1β and TNF-α mRNA between the Yaobitong, Yaotongning and Meloxieam groups (F= 3.26, P= 0. 086〉0.05). Conclusion.. Yaobitong Capsule increases the content of type II collagen of lumbar intervertebral discs, and down-regulates the mRNA expression of IL-1β and TNF-α, thus reduces production of some inflammatory mediators, which might be one of the mechanisms of Yaobitong Capsule treating lumbar disc herniation.
