微小RNA-200b对子宫内膜癌细胞中TIMP2基因表达的影响 被引量:3

摘要 微小RNA(microRNA,miRNA)是一类长约22个核苷酸的非编码小RNA分子,以转录后调节的方式改变靶基因的表达水平,参与细胞增殖、分化、凋亡等重要过程,在肿瘤发生发展、浸润转移中起重要作用.子宫内膜癌是威胁妇女健康的三大妇科恶性肿瘤之一,占女性生殖器官恶性肿瘤的20% ~ 30%,近年来发病率在各年龄层妇女中均呈上升趋势,并逐渐年轻化.目前,尚缺乏能够明确诊断而又能减少创伤性操作、更为有效的子宫内膜癌早期筛查诊断手段,影响着临床医师的决策.在前期研究中,本课题组已经发现子宫内膜腺癌与癌旁组织中111个miRNA表达存在差异,其中,差异最为显著的是上调表达的miRNA-200b(miR-200b)[1].为此,本研究通过荧光素酶报告基因检测、实时定量(QRT) PCR、蛋白印迹法(western blot)及ELISA方法,验证miR-200b是否直接抑制TIMP2基因的表达,为进一步研究miR-200家族的生物学功能,研究miRNA在肿瘤浸润转移过程中的分子机制提供实验支持,也为子宫内膜癌的预防和治疗提供新的手段.
