熊去氧胆酸与人血清白蛋白的相互作用研究

摘要 目的模拟正常人体生理条件下,采用光谱技术研究熊去氧胆酸与人血清白蛋白相互作用的机制。方法通过荧光光谱法和紫外光谱法确定熊去氧胆酸对人血清白蛋白荧光淬灭的机制,并通过Stern-volmer方程及双对数方程处理试验数据,得到两者结合常数和结合位点数。结果熊去氧胆酸在1.25×10-6-4.38×10-6 mol·L-1浓度范围内,对人血清白蛋白的内源荧光有较强的淬灭作用,淬灭类型属于静态荧光淬灭。熊去氧胆酸与人血清白蛋白的结合常数为5.08×102 L·mol-1,结合位点数为0.69。结论熊去氧胆酸与人血清白蛋白之间可形成稳定复合物,因此可以被白蛋白贮存、运载,通过血液循环达到作用部位。 Objective Under simulative physiological conditions, the interaction mechanism of human serum albumin (HSA) and ursodeoxycholic acid (UDCA) was studied by spectroscopy. Methods Fluorescence spectroscopy and ultraviolet spectroscopy were used to determine the fluorescence quenching mechanism of HSA caused by UDCA. Stern-volmer equation and double logarithmic equation were used to calculate various binding parameters. Results Fluorescence quenching of HSA by UDCA was strong in 1. 25 X 10^ 6 4. 38 X 10 ^6 tool · L1 range,and belongs to static quenching type. It was found that a complex formed between HSA and UDCA resulted in fluorescence quenching of HSA. The binding constant of the complex formed by UDCA and lISA was 5.08 X 10^2 L · mol 1 ,and the binding site number of UDCA to HSA was 0.69. Conclusion This study suggests that the UDCA can form stable complexes with HSA, which illustrats that UDCA can be stored and transported by HSA in the blood circulation.
