摘要
从裂冠牛奶菜(Marsdenia incisa P. T. Li et Y. H. Li)的粗甙酸水解产物中分离得到2个已知的C_(21)甾体甙元——肉珊瑚甙元(sarcostin, Ⅰ),青羊参甙元(qingyangshengenin,Ⅱ),和一个新的C_(21)甾体甙元,命名为裂冠牛奶菜甙元A(incisagenin-A,Ⅱ)经光谱分析和化学反应,证明(Ⅲ)的结构为12-β-氧-顺芷酰去酰基萝藦甙元。
From the acidic hydrolysate of crude glycoside of Marsdenia incisa P. T. Li et Y. H. Li, two known C21 steroidai aglyconcs-sarcostin ( I ) , qingyangshengenin (Ⅱ ) , and a new C21 steroidai aglyconc named incisagenin-A (Ⅲ) were isolated.
Compound (Ⅲ), colorless needle from acetone, mp 133-135 ℃, [α]D-14.2 (c =0.49, CHC13). On the bases of EI-MS and elemental analysis, give molecular formula :C26H38O7 (M+, 462). Compound (Ⅲ) liberated deacylmctaplexigenin (Ⅳ)
on alkaline hydrolysis. Its spectral signals-UV: 217 nm; IR: 1705, 1265cm-1;
MS: 101 (tiglic acid+ 1), 100 (tiglic acid), 83 (tigloyl cation, base peak), 55 tigloyl cation-CO), 1H NMR: 1.70 (1H, d, J=6.0Hz, 4'-Me) , l.98(3H, br.s, 5'-Me), 6.30(lH, br.s, 3'-H), 13C NMR: 166.5(C-l'), 129.4(C-2'), 126.8(C-3'), 14.2(C-4'), 12.3(C-5'). suggested the presence of tigloyl group. Comparing 1H NMR of (Ⅲ) and (Ⅳ), it was found that 5.5 (lH, dd, J =9.0, 6.0 Hz, 12-CHα) of (Ⅲ) upfields to 4.05 (lH, dd, J =9.0, 6.0 Hz, 12-CHα) of (Ⅳ). Therefore compound (Ⅲ) was established as 12-β-O-tigloyldeacylmctaple-xigenin, named incisagenin-A ( Ⅲ ) .
