寒凝证类痛经大鼠各时段子宫微循环的变化及不同针刺刺激量的干预效应 被引量:2

Effect of Different Quantity of Acupuncture Stimulus on Uterine Microcirculation in Dysmenorrhea Rats with Cold Syndrome at Different Time-points

摘要 目的:观察不同针刺刺激量对寒凝证类痛经大鼠各时段子宫微循环的影响,探讨刺激量与针刺效应之间的关系。方法:将动情间期的SD(Spraguc Dawlcy)雌性大鼠随机分为4组,分别是盐水组、模型组、刺激量A组(期望"得气"组)、刺激量B组(期望"不得气"组),每组8只。模型组和针刺组采用全身冷冻(-25℃环境下冷冻4h/天/5天)配合激素注射方法(每日给予大鼠皮下注射苯甲酸雌二醇注射液,连续10天,末次给药1h后,腹腔注射缩宫素2U/只),制备寒凝证类痛经大鼠模型。盐水组连续10日皮下注射等剂量的生理盐水,末次给药1h后,腹腔注射生理盐水2U/只。针刺组于第10日以不同刺激量针刺双侧"三阴交"穴20min。采用XW-B-3冷光源微循环显微检查仪观察各组大鼠子宫微循环在不同时段的变化。结果:与盐水组比较,模型组各时段的微血管管径、毛细血管管径(除40min、60min外)明显收缩(P<0.01)、流态减慢或停滞(P<0.01或P<0.05);与模型组比较,刺激量A组各时段的微血管管径、毛细血管管径(仅5min、10min、20min、30min)明显扩张(P<0.01或P<0.05),各时段流态明显改善(P<0.01或P<0.05)。刺激量B组微血管管径在30min出现明显扩张(P<0.01),微血管清晰度(仅50min、60min)有所改善(P<0.05);与刺激量B组比较,刺激量A组各时段的微血管管径,毛细血管管径(仅5min、10min、20min、30min)明显扩张(P<0.01或P<0.05),流态(除50min、60min外)明显改善(P<0.01或P<0.05)。结论:针刺可以改善子宫微循环障碍,不同针刺刺激量对子宫微循环的作用存在差异,刺激量A组针刺疗效显著优于刺激量B组,处于较强得气状态,间接说明了得气对针效的重要性。 Objective:To observe the effect of different quantity of acupuncture stimulus on uterine microcirculation in dysmenorrhea rats with cold syndrome at different time -points,and to discuss the relationship between quantity of acu-puncture stimulus and acupuncture effect .Methods:Dioestrum female rats were randomly divided into four groups , sa-line control group, model control group, A stimuli group(“deqi”group), B stimuli group(no“deqi”group), with eight rats in each group .Rats of model control group and stimulus groups were frozen in -25℃freezer for five days with four hours per day while given subcutaneous estradiol benzoate injection for ten days with one hour after the last injection , in-tra-peritoneal injection of oxytocin 2u/each.Saline control group was given the same dose of saline for ten days with one hour after the last injection , intra-peritoneal injection of saline 2u/each.On the tenth day, stimulus groups were given different quantity of acupuncture stimulus on both sides of “Sanyinjiao(SP6)” for 20 minutes.Use XW-B-3 Cold Light microcirculation tester to observe the changes of uterine microcirculation in each group at different time -points.Results:Model control group was compared with to saline control group , and diameters of micro -vessel at all time-points, diameters of capillary (except 40min and 60min) were significantly contracted (P〈0.01),blood flow slo-wer or stagnation (P〈0.01 or P〈0.05).Stimulus groups were compared with model control group , and about A stim-uli group, diameters of micro-vessel at all time -points, blood flow at all time -points was speeded up significantly (P〈0.01).About B stimuli group, diameters of micro -vessel were expanded only at 30min-point(P〈0.01).A stimuli group was compared with B stimuli group :diameters of micro -vessel at all time -points, diameters of capillary (only 5min,10min,20min and 30min)were significantly expanded (P〈0.01 or P〈0.05),blood flow(except 50min and 60min)were obviously improved(P〈0.01 or P〈0.05).Conclusion:Acupuncture could improve uterine microcir-culation.With different quantity of acupuncture stimulus , effects of uterine microcirculation were different between A stimuli group and B stimuli group ,the former was in a better and stronger“deqi”state,which showed the importance of“deqi”in acupuncture efficacy .
