摘要
目的:探讨RNA干扰技术沉默STAT3基因调控腺样囊性癌细胞生长的实验研究。方法:人腺样囊性癌生长细胞株ACC 2常规培养,分别采用A组为空白转染组,B组转染阴性对照siRNA(NC),C组转染特异性的siRNA(siRNA组)转染进ACC2,采用qRT PCR和Western blot法检测转染后STAT3 mRNA和蛋白表达水平的变化。MTT检测转染后腺样囊性癌细胞增殖。结果:ACC2细胞转染STAT3 siRNA 48 h后,STAT3 mRNA和蛋白的表达均受到明显抑制(P<0.05)。ACC 2生长72 h细胞转染后细胞增殖受到抑制,较对照组有显著性差异(P<0.05)。结论:STAT3基因有可能成为腺样囊性癌细胞生长基因治疗中重要的靶基因。
Objectives: To explore the effect of downregulation STAT3 expression on the growth of adenoid cystic carcinoma(ACC). Method: STAT3 siRNA were designed and chemical synthesized, then transfected into the human adenoid cystic carcinoma cell line, qRT- PCR and Western blot assay were used to detect STAT3 mRNA and protein expression level. MTT were used to detect proliferation in ACC2 cell. Result:STAT3 mRNA and protein expression in ACC2 cell transfected with siRNA STAT3 were inhibited significantly (P〈0.05) compared with the NC control group.Proliferation in ACC2 cell was inhibited significantly,and apoptosis was induced significantly compared with the NC control group (P〈0.05). Conclusion: STAT3 gene may be an important target gene in gene therapy to adenoid cystic carcinoma cancer.
