摘要
用RAPD技术和混合分离分析 (BSA)方法对双羔家系的 2 6只滩羊和对照系的 2 5只单羔滩羊进行了RAPD检测。发现 :OPB1 7-530、OPB1 9-92 6和OPB2 0 -1 4 2 6三条DNA片段仅出现在多羔家系的母本及多羔母本与单羔父本杂交产生的部分后代中 ,而在对单羔羊的逐个检测中未出现这三条片段。据此可以推断三个DNA分子标记与多羔性能主基因可能相关。
This study detected the genomic DNA polymorphisms in Tan sheep using RAPD technique with BSA(Bulked Segregant Analysis) to identify DNA markers unique to the fecund gene in Tan Sheep.Genomic DNA was isolated from leukocyte of 51 Tan sheep.One bulk contained mixed DNA samples of 26 indiveiduasl from prolific sheep,and another bulk was those of 25 individuals from general flock.The bulks were first screened for polymorphisms with 100 kinds of arbitrary 10 mers primers.Three polymorphisms in the high ovulation rate and litter size trait pool distinguished two bulks differently by primer OPB 17,OPB 19 and OPB 20.The size of those polymorphisms in 530bp、926bp and 1426bp.Futher screening using the 51 individual DNA sample indicated that the three polymorphic markers were present in 3 dams and 39%(9/23) hybrids that generated from mating of prolific dams with general size,but absent in each of the 25 Tan sheep from the general flock.The result suggested that the three markers,OPB17 530,OPB19 926 and OPB20 1426 might linked to the fecund gene in Tan sheep.
