摘要
本研究旨在探究氨磷汀(WR2721,Amifostine)对60Coγ射线6.5 Gy单次全身照射(TBI)所致急性放射病小鼠早期骨髓造血的防护作用。60只C57/BL6J小鼠随机分成正常对照(normal)、照射对照(IR)和WR2721预防给药(WR2721)3组。观察WR-2721照射前30 min给药对6.5 Gy60Coγ射线照射小鼠照射后60 d内小鼠外周血白细胞(WBC)、中性粒细胞(Neut)、血小板(Plt)和红细胞(RBC)数的影响;照射后2 h和24 h计数骨髓有核细胞数(BMNC)、流式细胞术分析造血干/祖细胞(LSK/LK)数量和观察多系骨髓细胞集落形成能力。结果显示,WR2721组的WBC和Neut在照射后4-18 d、Plt数7-18 d、RBC数10-30 d均明显高于IR组(P<0.05);照射后24 h BMNC、LSK和LK数均明显增加(P<0.05);2 h和24 h髓系祖细胞集落形成单位(CFU-GEMM)、粒系-巨噬系集落形成单位(CFU-GM)、巨核系集落形成单位(CFU-MK)、红系暴式集落形成单位(BFU-E)和红系集落形成单位(CFU-E)集落数均明显增加(P<0.01)。结论:WR2721能有效减轻急性放射病小鼠造血系统早期损伤,促进外周血细胞更早更快恢复。
The aim of this study was to investigate the effect of WR2721 (amifostine) against bone marrow hematopoietic damage of mice exposed to 6.5 Gy of 60Co γ-rays. A total of 60 C57/BL6J mice was divided into 3 groups: normal group (mice were injected with physiological salt solution), irradiation group (mice were injected with physiologic salt solution before irradiation) and WR2721 group ( mice were injected with WR2721 before irradiation). The WBC neutrophil (Neut), Pit and RBC levels in peripheral blood of 3 group mice were counted within 60 days after irradiation; the bone marrow nuclear ceils (BMNC) were counted at 2 and 24 hours after irradiation; the hematopoietic stem/progenitor cell (LK/LSK) level and colony formation capability were detected by flow cytometry at 2 and 24 hours after irradiatien. The results indicated that the counts of WBC and neut at 4 and 18 days, Pit at 7 - 18 days and RBC at 10 -30 day after irradiativn in WR2721 group were higher than those in irradiation group (P 〈0.05) ; the BMNC, LSK and LK levels obviously increased at 24 hours after irradiation ( P 〈 0.05 ), the CFU-GEMM 、 CFU-GM 、 CFU-MK BFU-E and CFU-E all significantly increased at 2 and 24 hours after irradiation( P 〈 0.01 ), as compared with irradiation group. It is concluded that WR2721 can effectively alleviate early hematopoietic damage and promote the fast recovery of peripheral blood cells in mice exposed to γ-ray, suggesting that the WR2721 has significant radiopretective effect on hematopoietic system.
