摘要
目的研究NAD+依赖性15-羟基前列腺素脱氢酶(15-PGDH)在食管癌组织及癌旁6 cm处非肿瘤对照组织中的表达,探讨15-PGDH在食管癌中的表达情况及其与食管癌的关系。方法本实验采用免疫组化SP法检测食管癌组织和癌旁6 cm处非肿瘤对照组织中15-PGDH的表达情况。结果免疫组化结果提示:40例食管癌组织中有5例(12.5%)15-PGDH呈强阳性染色,8例(20.0%)15-PGDH呈弱阳性染色,27例(67.5%)15-PGDH表达缺失;40例癌旁6 cm处对照组织中,13例(32.5%)15-PGDH呈强阳性染色,25例(62.5%)15-PGDH呈弱阳性染色,2例(5.0%)15-PGDH染色阴性(P<0.05)。15-PGDH在食管癌组织(32.5%)的阳性表达率显著低于癌旁6 cm处非肿瘤对照组织(95.0%)的阳性表达率,15-PGDH的表达差异有统计学意义(P<0.05)。15-PGDH蛋白表达与食管癌患者的性别、年龄、分化程度无关(P>0.05)。15-PGDH蛋白表达与食管癌患者有无淋巴结转移有关(P<0.05)。结论 15-PGDH在食管癌组织中表达缺失或减少,15-PGDH可能是食管癌发生发展中的重要抑制因子。
Objective To study the expression of NAD^+-dependent 15-hydroxyprostaglandin dehydrogenase (15-PGDH) in the tissue of esophageal carcinoma and normal esophageal mucosa next to the carcinoma,and investigate its possible implication.Methods In the tissue of normal esophageal mucosa next to the carcinoma (40 cases),esophageal carcinoma (40 cases),the protein expression of 15-PGDH was detected by immunohistochemistry SP method.Results Immunohistochemistry:among 40 cases of esophageal carcinoma,there was no expression of 15-PGDH in 27 cases (67.5%),8 cases (20.0%) showed a weak positive staining of 15-PGDH,5 cases (12.5%) of 15-PGDH was strongly positive staining; among 40 cases of normal esophageal mucosa next to the carcinoma,13 cases (32.5%) of 15-PGDH was strongly positive staining,25 cases (62.5%) showed a weak positive staining of 15-PGDH,there was no expression of 15-PGDH in 2 cases (5.0%) (P 〈 0.05),15-PGDH expression was significantly different between two groups (P 〈 0.05).Immunohistochemical results suggested that the expression rate of 15-PGDH in normal esophageal mucosa next to the carcinoma was significantly higher than that in esophageal carcinoma and the difference had a statistically significance (P 〈 0.05).The low expression of 15-PGDH in esophageal carcinoma was found to be unrelated to the histological differentiation and the increased stages (P 〉 0.05).Conclusion In the tissue of normal esophageal mucosa next to the carcinoma,esophageal carcinoma,15-PGDH protein expression is decreased gradually.15-PGDH is correlation to the esophageal canceration and growth.The expression of 15-PGDH is absent or decreased in esophageal carcinoma.The loss or reduction of 15-PGDH may occur in the development of esophageal carcinoma.15-PGDH is an inhibitor of esophageal tumor genesis.
