摘要 目的探讨三氧化二砷(As2O3)对增生性瘢痕成纤维细胞(HSFB)增殖和胶原合成的影响及其意义。方法通过MTT比色试验和生长曲线测定法检测As2O3对体外培养的HSFB增殖活性的影响;3H-脯氨酸掺入法测定As2O3对细胞胶原蛋白合成的影响;用光镜和电镜观察药物作用后HSFB形态学和超微结构的改变。结果 As2O3明显抑制了HSFB的增殖和胶原蛋白的合成,且在一定范围内具有时间-剂量依赖性,以8μg/L浓度组第3天作用最为显著;HSFB的形态和超微结构呈抑制改变。结论As2O3具有抑制HSFB增殖和胶原合成的作用,为应用该药治疗瘢痕提供了实验依据。 Objective To study the effect of As2O3 on proliferation and collagen synthesis of hypertrophic sear fibroblasts and its significance. Methods Fibroblasts derived from human hypertrophic sear were cultured in vitro, the influence of As2O3 on cell proliferation and collagen synthesis of the fibroblasts were investigated by MTf eolorimetrie assay, cell growth curve and ^3 H-proline uptake analysis. Light microscope (LM) and electron microscope (EM) were employed to study the morphology and uhrastructure changes in fibroblasts after the application of As2O3. Results In definite range, As2O3 markedly inhibited the proliferation and collagen synthesis of HSFB in a time-dose manner, especially treated with 8 μg/L concentration at 3 days later. As2O3 obviously restrained the morphology and ultrastructure of fibroblasts. Conclusion As2O3 can significantly inhibit the proliferation and collagen synthesis of HSFB, which may provide some foundation for treating pathological scar.
