缺血性脑卒中火毒证病证结合动物模型的研究 被引量:15

Establishment of disease-syndrome combination animal model of ischemia stroke-heat toxin syndrome

摘要 目的探索建立缺血性脑卒中火毒证病证结合大鼠模型的制备方法和评价体系。方法通过腹腔注射角叉菜胶复合线栓大脑中动脉阻塞法(MCAO)建立缺血性脑卒中火毒证大鼠模型。观察模型组大鼠在不同时间点3、6、24、72 h和7 d的舌部、耳廓、爪甲、尾部的表征,检测体温、相关凝血指标、脑组织形态学等方面的变化,建立该模型的评价体系。结果腹腔注射角叉菜胶后,模型大鼠出现舌色发暗、舌下络脉增粗增长、耳廓边缘发红、爪甲肿胀红紫、尾部颜色暗红等瘀热表征,复合MCAO法后模型大鼠瘀热表征较假手术组更明显,呈持续性加重;且体温在造模24 h后有大幅度升高,至造模72 h仍高于假手术组,接近于中医的火毒征象;凝血指标提示模型组在不同时间点全血黏度增高的同时血液处于低凝状态,脑组织形态学符合缺血性改变。结论角叉菜胶复合MCAO可以制备缺血性脑卒中火毒证大鼠模型;表征、体温、凝血指标、脑组织形态学的综合评价可构成该模型的评价体系。 Objectives To explore the preparing method and reviewing system for establishing diseasesyndrome combination rat model of ischemia stroke-heat toxin syndrome. Methods The rat model of ischemia stroke-heat toxin syndrome was established with intraperitoneal injection of carrageenan combining thread approach of middle cerebral artery occlusion ( MCAO ). The representations of rat tongue, auricle, nail and, tail, and changes of body temperature, relative coagulation indexes and cerebral morphology were observed and detected at different time points for establishing a review system. Results The model rats showed some stasis-heat representations including black tongue, thickened sublingual collateral vessels, red auricle, swelling and purple nail and dark tail after the intraperitoneal injection of carrageenan. After MCAO, the representations were more significant in model group than those in sham-operation group and exacerbated persistently. The body temperature increased sharply after 24 h, and was still higher in model group than that in sham-operation group after 72 h, which was similar to TCM heat toxin syndrome. The coagulation indexes showed that whole blood viscosity increased but blood was low coagulation state in model group at different time points. The changes of cerebralmorphology were accorded with ischemia changes. Conclusion The intraperitoneal injection of earrageenan combining thread approach of MCAO can be used for establishing rat model of ischemia stroke-heat toxin syndrome. The review system of the model can be consisted of representations, body temperature, coagulation indexed and changes of cerebral morphology
