补肾生血解毒方对再生障碍性贫血小鼠红细胞生成相关细胞因子表达的影响 被引量:11

Influences of Bushen Shengxue Jiedu Fang on expressions of erythropoiesisrelated factors in mice with aplastic anemia

摘要 目的探讨补肾生血解毒方对理化复合因素致骨髓抑制小鼠红细胞生成的影响。方法 50只BALB/C小鼠随机分为空白组、模型组、中药小、中、大剂量组,以^(60)Co-γ射线复合环磷酰胺致小鼠骨髓造血抑制为再生障碍性贫血(AA)模型,补肾生血解毒方(龟板胶、鹿角胶、人参、枸杞子、黄芪、当归、黄连)干预,中药小、中、大剂量组分别给予相当于生药量6.14、12.29和24.57 g/kg灌胃。血细胞计数仪检测小鼠外周血中红系细胞各项参数的改变,酶联免疫吸附测定法(ELISA)检测外周血血清中红细胞生成素(EPO)、干扰素γ(IFN_γ)含量的差异,RT-PCR检测骨髓细胞中EPO、IFN_γ受体(IFN_γR1)基因表达的改变。结果补肾生血解毒方可以明显提高AA小鼠外周血红系细胞参数,缓解贫血症状,显著增加外周血血清中EPO含量和骨髓细胞中红细胞生成素受体(EPOR)基因的表达,并可以降低外周血中IFN_γ的含量,抑制IFN_γR1基因的表达。结论补肾生血解毒方治疗再生障碍性贫血的机制可能与提高EPO活性,并抑制由IFN_γ介导的免疫反应,恢复造血干细胞的正常功能,恢复红细胞的正常分化和成熟有关。 Objective To investigate the influence of Bushen Shengxue Jiedu Fang on erythropoiesis in myelosuppression mice induced by physical and chemical compound factors. Methods BALB/C mice (n = 50) were randomly divided into blank group, model group, and low-dose, mid-dose and high-dose Chinese medicinal groups ( low-dose group, mid-dose group and high-dose group). The model of aplastic anemia (AA) was established with 60Coγ ray and cyclophosphamide injection. Bushen Shengxue Jiedu Fang was used for intervention in low-dose, mid-dose and high-dose groups (6.14 g/kg, 12.29 g/kg and 24.57 g/kg). The changes of erythrocyte parameters were detected by using blood cell counter. The level changes of erythrogenin (EPO) and interferon γ (IFNγ) were detected by using ELISA, and expressions of EPO receptor and IFNγ receptor were detected by using RT-PCR. Results BushenShengxue Jiedu Fang increased erythrocyte parameters, relieved anemia symptoms, increased the level of serum EPO and improved significantly the expressions of EPO receptor gene in bone marrow cells, decreased the content of IFNγ in peripheral blood, and inhibited the expression of IFNγ receptor gene. Conclusion Bushen Shengxue Jiedu Fang may have curative effect on AA through increasing EPO activity, inhibiting immune reaction induced by IFNγ, recovering normal function of hematopoietic stem cell, and promoting normal differentiation and mature of red blood cells.
