转融合基因Cry1Ab/Ac水稻稻杆对泥鳅蛋白水解酶和乙酰胆碱酯酶活性的影响 被引量:1

摘要 为探索转Bt抗虫水稻对泥鳅的毒性作用,采用水体中添加不同浓度(10、50、100、200 mg/L)的转融合基因Cry1Ab/Ac水稻华恢1号(HH1)稻杆粉饲养泥鳅,非转基因亲本明恢63(MH63)作对照,不加稻杆粉为空白对照,分析了对泥鳅蛋白水解酶、乙酰胆碱酯酶(AChE)的影响。研究发现:经4个浓度处理后,泥鳅的胃蛋白水解酶、肠蛋白水解酶和脑AChE、肝脏AChE、肌肉AChE与对照组泥鳅相比均无显著差异(n=3,P>0.05)。高浓度(200 mg/L)HH1、MH63组泥鳅胃蛋白水解酶与空白对照相比均显著降低(n=3,P<0.05);低浓度(10 mg/L)HH1组泥鳅肝脏AChE与空白对照差异显著(n=3,P<0.05);不同浓度HH1、MH63稻杆粉对泥鳅的肠蛋白水解酶、脑AChE和肌肉AChE均无显著影响(n=3,P>0.05)。结果表明:HH1水稻稻杆粉对泥鳅的消化道蛋白酶和乙酰胆碱酯酶没有影响,而水体中高剂量的HH1或MH63稻杆粉均会降低泥鳅胃的消化能力。 In order to explore the effects of transgenic Bt rice on the activity of proteolytic enzyme and acetylcholinesterase(AChE), Misgurnus anguillicaudatus Were fed in 4 levels(10,50,100 and 200 mg/L)in water which formulated by rice straw powder. Added transgenic Cry1Ab/Ac rice (HH1) straw powder was test group, and near-isogenic non-Bt rice (MH63) straw powder as a control, without rice straw powder as control check. The results showed that stomach proteolytic enzyme, intestines proteolytic enzyme, brain ACHE, liver AChE and muscle AChE of M.anguillicaudatus were raised with HH1 rice straw powder had no significant differences compared to that of M.anguillicaudatus fed with MH63 straw powder (n=3,P〉0.05). However, stomach proteolytic enzyme of M.anguillicaudatus fed with 200 mg/L HH1 straw powder were lower than that M.anguillicaudatus of control check (n=3 ,P〈0.05), and the stomach proteolytic enzyme of M. anguillic audatus of MH63 group reduced significantly than that of control check too (n=3 ,P〈0.05). Compared with M.anguillicaudatus of control check, the activity of liver AChE of that of 10 mg/L HH1 test group showed obviously significant different(n=3 ,P〈0.05). But intestines proteolytic enzyme, brain AChE and muscle AChE of M.anguillicaudatus were not affected by adding different concentration of rice straw powder to water(n=3 ,P〉0.05). In conclusion, transgenic Cry1Ab/Ac rice (HH1) straw powder could have no direct negative impact on the activity of proteolytic enzyme and acetylcholinesterase of M.anguillicaudatus. But the higher concentration of rice straw powder could influence on the proteolytic enzyme of M.anguillicaudatus.
