摘要
研究了对 β 硫酸酯乙砜基苯胺活化法在棉织物上固定化脂肪酶的工艺条件 ,并考察了固定化酶的最适作用温度和pH及间歇操作稳定性。最佳固定化条件为 :醚化pH为 10 0 ,偶联pH为 7 0 ,脂肪酶的浓度为 6mg ml,偶联时间为 12h ,所得固定化脂肪酶的最大活力为 35U g(棉 )·min。固定化酶的最适作用温度为 35℃ ,最适pH为 8 0 ,半衰期为 6d。
The conditions of the lipase immobilization onto cotton fiber activated by p\|β\|sulfuric acid ester ethyl sulfone aniline (SESA) have been studied.The optimal temperature and pH of the immobilized lipase were investigated,and the operating stability of the immobilized lipase was studied too.The results showed that the optimal conditions for the immobilization were that the pH of the reaction for the cotton fiber with SESA was 10.0,the linking pH of lipase to fiber was 7.0,the linking time was 12h,and the concentration of lipase was 6mg·mL -1 .Then 35 U·g(fiber) -1 ·min -1 activity of immobilized lipase was obtained. The optimal temperature and pH of the immobilized lipase were 35℃ and 8.0 respectively,and its half life was 6 days.
