摘要
对红酵母RY-98细胞壁破碎及其类胡萝卜素浸提的方法进行了较详细的筛选和条件优化研究。结果表明:酸热法是红酵母破壁较理想的方法,其最优条件组合为盐酸浓度2 mol/L、盐酸用量12mL/g菌体、盐酸浸泡时间40 min和沸水浴处理4 min;丙酮是从红酵母细胞碎片浸提类胡萝卜素较好的有机溶刺,其参考用量为20 ml/g菌体。在此破壁和浸提条件下,红酵母RY-98类胡萝卜素提取率可达450.2μg/g菌体,比优化前提高37.1%。
The methods of breaking cell walls and extracting carotenoids were studied. The experimental results showed that the method of combining hydrochloric acid (HCL) with heating (by boiling water) was suitable for the cell rupturing from Rhodorula RY - 98, and its optimal conditions were as follows: HCL concentration 2 mol/L, HCL addition 12 ml/g (dry cells), soaking time 40 min and heating time 4 min. Using acetone of 20 mL/g (dry cells) was the most extracting method. Under the above optimal conditions, the yield of carotenoids could reached 450.2 μg/g (dry cells).
