摘要
为了研究FSHβ与FSHR基因在小梅山猪不同生长阶段表达规律,试验采用实时荧光定量PCR(RQ-PCR)方法分别对初生、1月龄、2月龄、3月龄、4月龄、5月龄小梅山猪的FSHβ与FSHR基因表达水平进行检测。结果表明:小梅山猪从初生到5月龄下丘脑-垂体-性腺轴下各组织中均检测到FSHβ与FSHR基因的表达;不同组织中,FSHβ基因在垂体中均高度表达,输卵管和卵巢则低度表达,4月龄时,FSHβ基因在垂体中的相对表达量是输卵管的54.557倍;FSHβ的表达水平在不同生长阶段中呈现先上升后下降趋势,4月龄时达到高峰;4月龄前,FSHR基因在下丘脑中高度表达,4月龄之后,则卵巢和下丘脑均为高度表达,而子宫均为低度表达。说明小梅山猪的初情期在4月龄左右。
To study the expression patterns of FSHβ and FSHR genes in Xiaomeishan pig during different growth stages, the method of real - time fluorescence quantitative PCR ( RQ - PCR) was used to detect the expression levels of FSHβ and FSHR genes of Xiaomeishan pig at birth, one month old, two month old, three month old, four month old and five month old, respectively. The results showed that the expressions of FSHβ and FSHR mRNA could be detected in various tissues of lhe hypothalamic - pituitary - gonadal (HPG) axis in Xiaomeishan pig from birth to five month old. The FSHβ genes were highly expressed in pituitary and lowly expressed in oviduct and ovary in the different tissues. The relative expression level of the FSHβ genes in pituitary was 54. 557 times higher than that in oviduct for Xiaomeishan pig at four month old, and the expression level of the FSHβ genes showed an increasing trend at first, followed by a decreasing trend during different growth stages, which reached the peak at four month old. The FSHR genes were highly expressed in hypothalamus before four month old, and then were highly expressed in ovary and hypothalamus, were lowly expressed in uterus after four month old. The results indicate that the puberty in Xiaomeishan pig is at four month old or so.
