云南半细毛羊微卫星标记与羊毛纤维直径相关性研究

The Study on the Correlation of Microsatellite DNA Markers With Wool Fiber Diameter in Yunnan Semi-fine wool Sheep

摘要 应用10个微卫星标记对99个云南半细毛羊基因组DNA进行多态性分析,并结合样本的羊毛纤维直径,利用SAS软件进行了最小二乘相关性分析及多重比较,以期寻找对云南半细毛羊羊毛纤维直径有显著影响的分子标记及其基因型,为云南半细毛羊的保存、开发、利用提供科学、客观的理论依据。结果表明:云南半细毛羊10个微卫星位点PIC值均大于0.5,表明遗传多样性都极为丰富;微卫星位点BMS1714及BM6404对云南半毛羊的羊毛纤维直径有极显著影响(P<0.01),位点BMS1248对该性状具有显著影响;位点BMS1714可检出7个等位基因,11种基因型,其中基因型141/141对羊毛纤维直径具有正效应;位点BMS1248可检出4个等位基因,6种基因型,其中基因型135/145对羊毛纤维直径具有负效应;位点BM6404可检出10个等位基因,9种基因型,其中基因型141/145的最小二乘均数最大,对羊毛纤维直径具有正效应。在云南半细毛羊的纤维直径选择中,微卫星位点BMS1714、BMS1248有可能作为云南半细毛羊纤维直径选择的参考分子标记,而位点BM6404在以往的研究中未见与纤维直径相关报道,所以能否作为该性状的参考标记还需深入研究。 Ten micros atellites loci were studied to detect their genotypes in 99 individuals in a Yunnan Semi-fine Wool Sheep. In order to find the markers which are significantly correlated with the phenotype of wool fiber diameter, A SAS procedure was used to analyze the effects of these 10 microsatellites on wool fiber diameter. The result showed the PIC value of Yunnan semi-fine wool sheep was more than 0.5, indicating more abundant genetic diversity. The result of correlative analysis between microsatellite markers and phenotype indicated that the locus BMS1714 and BM6404 had extremely significant (P〈0.01) correlation with wool diameter. The locus BMS1248 had significant (P〈 0.05) correlation with wool diameter. In the marker BMS1714, geuotype 141/141 had positive effect on wool fiber diameter. In the marker BM6404, genotype 141/145 had positive effect on wool fiber diameter. In the marker BMS1248, genotype 135/145 had negative effect on wool fiber diameter. The above results indicated marker BMS1714 and BMSl248 may be used in Yunnan semi-fine wool sheep breeding. Further research is required on relationship between locus BM6404 and fiber diameter.
