姜黄素对人骨肉瘤MG63细胞凋亡的影响

摘要 目的探讨姜黄素诱导人骨肉瘤细胞株MG63凋亡的机制。方法不同浓度(0、25、50和75μM)姜黄素作用MG63细胞24 h后,CCK8法检测细胞体外增殖,TUNEL染色法检测细胞凋亡,Western blotting检测AKT/Bax-Bcl-2/Caspase 3等内源性凋亡途径相关蛋白表达水平。结果 CCK8实验结果显示在0~75μM范围内,50μM以上浓度的姜黄素能显著抑制MG63增殖,诱导细胞凋亡、p-AKT蛋白水平明显降低,而Bax蛋白及剪切的Caspase 3蛋白水平则显著增高。结论姜黄素通过影响AKT/BaxBcl-2/Caspase 3途径诱导MG63细胞凋亡。 Objective To study the effect of curcumin on the apoptosis of osteosarcoma cell line MG63.Methods After MG63 cells were treated with different concentration(0、25、50 and 75 μM) curcumin for 24 h,CCK8 assay was used to evaluate the proliferation and TUNEL assay was used to investigate the apoptosis of MG63 cells.Western blotting was performed to detect the expression level of p-AKT,Bax,Bcl-2 and Caspase 3.Results During the concentration of 0 to 75 μM,curcumin above 50 μM could inhibit the proliferation,induce the apoptosis of MG63 cells,and could inhibit the phosphorylation of AKT protein,while upregulate the expression of Bax and cleaved 3.Conclusion Curcumin could induce MG63 cells apoptosis by regulating AKT/Bax-Bcl-2/Caspase 3 pathway.
