摘要
为探讨优化黑线姬鼠-小鼠异种核移植技术体系,采用Piezo辅助胞质内体细胞核注射的方法进行克隆胚胎的重构,比较了不同供体细胞来源.不同浓度TSA以及不同供体细胞传代次数对重构胚的卵裂率和囊胚率产生的影响.结果表明,同种核移植试验中获得重构胚的卵裂率、囊胚率均显著高于异种核移植试验(P<0.05);传15代次的细胞作为供体细胞进行核移植,所得异种重构胚的囊胚率显著低于其他组别(P<0.05);50nmol/L和80nmol/LTSA处理组的囊胚率分别为6.06%和6.31%,显著高于其他各处理组及对照组的囊胚率3.13%(P<0.05).
In order to optimize the inter-species nuclear transfer system of Apodemus agrarius and mouse,we reconstructed embryos with the method of Piezo-assisted intracytoplasmic nuclear injection,and compared the different sources of donor cells,different concentrations of TSA,and effects of different donor cell passages act on cleavage rate and blast cyst rate of the reconstructed embryo.The results show that cleavage rate and blast cyst rate which were obtained in the same nuclear transplantation were significantly higher than those of inter-species nuclear transfer test(P〈0.05).The 15generation cells as donor cells for nuclear transfer,the interspecies reconstructed embryo blast cyst rate from the test was significantly lower than other groups(P〈0.05).The blast cyst rate of treatment group in 50and 80nmol·L^-1 TSA are 6.06%and 6.31%,significantly higher than the blast cyst rate 3.13%in the other treatment group and the control group.
