摘要
目的研究糖肾宁对糖尿病肾病(DKD)肾组织AT1R及细胞因子的影响,探讨其改善肾脏血流动力学的机制。方法采用雄性SD大鼠予高脂饲料诱导加腹腔注射链脲佐菌素(STZ)复制糖尿病肾病大鼠模型。造模成功大鼠40只,随机分为5组(每组8只):模型组(M组)、糖肾宁组(T组)、糖肾宁+格列喹酮组(T+G组)、格列喹酮组(G组)、福辛普利+格列喹酮组(F+G组),并给予相应药物干预。正常对照组(N组)10只大鼠给予普通饲料喂养,腹腔注射等量枸橼酸缓冲液。干预8周后,检测各组大鼠肾组织AT1R、VEGF、IGF-1和TGF-β表达。结果 M组AT1R、VEGF、IGF-1和TGF-β表达增高,与N组比较有显著性差异(P<0.01);各药物干预组AT1R、VEGF、IGF-1和TGF-β表达均低于M组(P<0.05,P<0.01)。结论糖肾宁独立于降压和降糖药物抑制肾组织AT1R、VEGF、IGF-1和TGF-β表达,从而改善肾脏血流动力学以保护肾脏。
Objective To observe the effect of "Tangshenning Recipe" on AT1R and cytokines in renal tissues, and to investigate its mechanism of renal hemodynamics. Methods Male SD rats were fed with high-lipid food along with injecting of streptozotoein (STZ) to depulicate the DN rat models. Forty rat models were randomly divided into 5 groups : model group ( group M), "Tangshenning Recipe" group ( group T), Tangshenning + Gliquidone ( group T + G), Gliquidone ( group G), Fosinopri + Gliquidone ( group F + G), with 8 rats in each group. Each group was treated with con'esponding therapies. Ten rats in normal group (group N) were fed with conventional diet and intraperitoneally injected with the same dosage of citrate buffer. After 8 weeks, the expressions of AT1R, VEGF, IGF-1 and TGF-[3 were detected. Results The expressions of AT1 R, VEGF, IGF-! and TGF-[3 in M group were significantly increased, with significant difference with that in N group ( P 〈 0.01 ) ; the expressions of AT1 R, VEGF, IGF- 1 and TGF-β in drug intervention groups were lower than that of the M group ( P 〈 0.05, P 〈 0.01 ). Conclusion "Tangshenning Recipe" can protect kidney and improve the kidney hemodynamics by inhibiting expressions of AT1R, VEGF, IGF-1 and TGF-β in renal tissues.
