复合酶法提取人参总皂苷 被引量:6

摘要 目的在人参根及茎叶总皂苷提取工艺中引入酶因素,提高人参总皂苷提取率。方法以人参总皂苷提取率为指标,运用单因素实验及正交试验确定复合酶作用的最佳工艺。结果无酶条件下人参根中总皂苷的提取率为3.22%,复合酶辅助法提取人参根总皂苷的最佳工艺条件为:中温淀粉酶5%,中性蛋白酶1%,时间2 h,温度60℃,得提取率为5.44%;无酶条件下人参茎叶中总皂苷的提取率为5.85%,复合酶辅助法提取人参茎叶总皂苷的最佳工艺条件为:漆酶5%,纤维素酶5%,甘露聚糖酶1%,时间2 h,温度40℃,提取率为11.93%。结论复合酶辅助提取法能够显著提高人参总皂苷提取率,且工艺简单、稳定性好,可用于产业化生产。 Objective To improve the extraction rate of ginsenosides by introducing enzyme factors in ginseng root and saponin during its extraction process. Methods Took the extraction rate of total ginsenoside as the indicators, and the optimal process was determined by single-factor experiments and orthogonal test. Results The resuhes of the experiment showed that the extraction rate of total saponins in ginseng root was 3.22%. The optimum extraction conditions of ginseng saponins by enzymatic method was the temperature amylase dosage was 1%, the neutral protease dosage was 1%, hydrolysis time was 2h, hydrolysis temperature was 60℃, extraction rate was 5.44%. The traditional process of total saponins extracted from ginseng stem leaf extraction yield of 5.85%. The optimum extraction conditions of ginseng stems and leaves saponins by enzymatic method was the laccase dosage was 5%, the cellulose dosage was 5%, the mannanase dosage was 5%, hydrolysis time was 2h, hydrolysis temperature was 40℃, extraction rate was 11.93%. Conclusion Enzyme-assisted method is efficient, simple operation and stable, which can be used for industrial production.
