摘要
为了建立野生稻的脱分化与分化体系,以普通野生稻为试验材料,通过在N6D、NB基本培养基中添加不同质量浓度激素(IAA、NAA、2,4-D、6-BA、KT),统计野生稻在不同诱导培养基上的出愈率,从而确定最佳诱导培养基,并观察胚性愈伤组织分化成苗的情况。结果发现,以N6D为基本培养基,添加质量浓度分别为6.0mg/L、3.0mg/L、3.0mg/L、1.5mg/L的2,4-D、NAA、IAA、KT,接种7d后出愈率高达56.7%,愈伤组织直径为0.31cm,将其转入MS分化培养基,24d后分化成苗。说明该诱导培养基是培养普通野生稻产生胚性愈伤组织的最佳培养基。
The N6D and NB medium containing different hormones(IAA, NAA, 2,4 -D, 6 -BA and KT)were used to study the callus rate of Oryza rufipogon Grill,so as to determine the best medium for O. rufipogon Grill. And the shoot induction rate from embryonic calli was also observed. The result showed that,after the seeds were placed on medium for 7d,the callus ratio from N6Dmedium with 6.0 mg/L 2,4- D,3.0 mg/L NAA, 3.0 mg/L IAA, 1.5 mg/L KT was 56.7% ,while the diameter of callus reached to 0.31 em. And then the callus of O. rufipogon Grill were transferred to MS dedifferentiation medium for 24 d, the callus became the green plants. This is a quick system for differentiation and dedifferentiation of O. rufipogon Griff.
