摘要
为研究半枝莲多糖对小鼠免疫功能的影响,探讨其可能的抗肿瘤作用机制,分别以50、100、200mg/kg的剂量对荷瘤小鼠进行肌肉注射,采用酸性α-醋酸萘酯酶(ANAE)染色法检测T淋巴细胞含量,利用流式细胞术检测T淋巴细胞亚群变化及表面蛋白CD28的表达,分别用IL-2、IFN-γ定量酶联免疫检测试剂盒测定IL-2、IFN-γ含量。结果显示,与对照组相比,半枝莲多糖各剂量组外周血T淋巴细胞总数均升高,中、高剂量组T淋巴细胞含量分别显著升高21.75%、26.44%;外周血T淋巴细胞亚群中的CD4+T细胞、CD8+T淋巴细胞均提高,CD4+/CD8+值增加,中、高剂量组CD4+T淋巴细胞含量分别显著提高6.53、4.92个百分点;各剂量组CD8+CD28+T淋巴细胞含量均提高,中、高剂量组分别极显著提高8.24、7.32个百分点,CD8+CD28-T淋巴细胞含量均低于空白对照组;各剂量组外周血血清中IL-2和IFN-γ的含量均提高,中剂量组分别提高100.30%、376.95%。可见,半枝莲多糖能够提高T细胞含量,提高CD4+/CD8+值,促进外周血血清中IL-2和IFN-γ的分泌,从而提高机体的免疫功能使其趋于稳态,发挥抗肿瘤作用。
To investigate the regulatory effects of Scutellaria barbata polysaccharides(SPS) on the immune function and anti-tumor mechanism in mice. SPS with the concentration 50,100,200 mg/mL was injected into the muscle of mice. T lymphocyte percentage was tested with ANAE and the change of T lymphocytes subsets and the expression of surface protein CD28 were tested with flow cytometry. IL-2 and IFN-γ, concentration were tested with ELISA kit. Compared with control group, the total number of peripheral blood T lymphocytes in SPS treatment group increased,and the middle and high group significant increased 21.75% and 26.44% ,respectively. The number of CD4^+T lymphocytes, CD8^+ T lymphocytes and the ratio of CD4^+/CD8^+ of lymphocyte subsets in SPS treatment group of T lymphocytes subsets also increased, and the middle and high group significant increased 6.53 and 4.92 percenge points, respectively. The number of CD8^+ CD28^+ T lymph cells in SPS treatment group increased,and the middle and high group significant increased 8. 24 and 7.32 percenge points, while the number of CD8^+ CD28^- T lymphocytes decreased. IL-2 and IFN-γ concentration increased in SPS treatment group, and the middle group increased 100.30% and 376.95% ,respectively. SPS improved the mice immunologic and anti-tumor function by promoting the proliferation of mice T lymphocytes,raised the ratio of CD4^+/CD8^+ of lymphocyte subsets,promoting the secretion of IL-2 and IFN-γ.
