食源性肥胖小鼠胰岛素抵抗模型的建立 被引量:5

摘要 本试验通过改良的高糖高脂饲料饮食诱导建立肥胖和胰岛素抵抗动物模型。我们将40只雄性C57BL/6J小鼠随机分为肥胖诱导组(DIO组)和正常对照组(CHOW组),分别喂以高糖高脂饲料和基础饲料4个月后,观察它们的体重和血糖情况,发现两组小鼠无论在体重还是血糖水平上差异都极其显著;对两组小鼠做葡萄糖耐量试验和胰岛素耐受试验,发现DIO组小鼠表现出明显的糖耐量异常和胰岛素抵抗现象;最后检测血清胰岛素水平发现,DIO组小鼠与CHOW组比较出现了明显的高胰岛素症状。故改良的高糖高脂饲料饮食诱导是一种经济实用、快速的肥胖和胰岛素抵抗动物模型建立的方法。 Diet--induced obese and insulin resistance mouse model is an essential tool for the study of human obesity and diabetes. We evaluated the effect of a homemade high-- fat diet in the induction of obesity in a mouse model. Male C57BL/6J mice were randomly divided into high fat group (DIO group) and normal group (CHOW group). When fed with the high sugar and high fat for 4 months, mice had significantly enhanced body weights, liver and fat tissue weights, and elevated blood glucose levels. In addition, impaired glucose tolerance and insulin resistance were observed during the glucose and insulin tolerance tests. Thus, with our homemade high fat diet, we have successfully generated a mouse model of obesity and insulin resistance with very low cost. Such model is critical for the study of obesity and diabetes.
