Detection of telomerase activity by PCR-ELISA in exfoliated cells in urine of bladder carcinoma patients

摘要 目的 探讨尿脱落细胞端粒酶活性变化在膀胱肿瘤诊断中的作用。 方法 应用PCR ELISA法检测 5 3例膀胱肿瘤患者尿液脱落细胞端粒酶活性。 结果 非膀胱肿瘤和膀胱肿瘤患者尿液脱落细胞端粒酶活性阳性率分别为 6 4.15 % (34/ 5 3)和 7.6 9% (2 / 2 6 ) ,健康对照者 7例均为阴性 ,膀胱肿瘤患者与正常人及非膀胱肿瘤患者的端粒酶活性分别相比 ,差别均有极显著性意义 (P<0 .0 0 1)。但端粒酶活性与肿瘤的分期分级无相关性。 结论 尿脱落细胞端粒酶活性检测可以作为诊断膀胱肿瘤的无创性检测方法 。 Objective To assess the telomerase activity of exfoliated cells in urine so as to find a new early noninvasive assay for the early diagnosis of bladder carcinoma Methods Telomerase activity in exfoliated cells in urine was detected by polymerase chain reaction enzyme linked immunosorbent assay (PCR ELISA) in 53 bladder carcinoma patients and in 26 non neoplastic bladder lesions and 7 normal healthy volunteers as controls. Results No telomerase activity was detected in urine samples of normal healthy volunteers.Telomerase activity was detected in urine exfoliated cells in 34 of the 53 (64.15%) bladder cancer patients.Whereas,only 2 of 26 urine samples from nonmalignant bladder diseases revealed a weak telomerase activity (7.69%).No association could be detected between telomerase activity and tumor stage or tumor grade. Conclusions PCR ELISA assay of telomerase activity in exfoliatd cells in urine is an early noninvasives means for the early diagnosis of bladder carcinoma however it is not associated with the tumor stage or tumor grade.
