大鼠小肠移植排斥反应中细胞黏附分子的动态观察

摘要 目的:研究细胞间黏附分子(ICAM-1)与大鼠小肠移植排斥反应的关系,以及雷帕霉素(RAPA)对其作用。方法:以SD大鼠为假移植组作对照,采用Wistar→SD大鼠异位小肠移植模型,根据不同处理分为排斥组、雷帕霉素2 mg/kg组和雷帕霉素4 mg/kg组,术后1、3、5、7天获取移植肠组织行病理学检查和ICAM-1免疫组化检测。结果:术后1、3天,各组均未发现排斥反应。术后5、7天,2组分别出现Ⅰ度和Ⅱ度排斥,3组于术后7天仅有不典型的排斥迹象,4组未见排斥证据。ICAM-1在对照组仅在血管内皮细胞和粘膜固有层有少量表达,在排斥组术后3天在上述部位表达增加,于第5天到达高峰。淋巴细胞和间质细胞表达明显增高,与对照组相比有显著性差异。RAPA4 mg/kg组能显著抑制移植后ICAM-1的表达。结论:ICAM-1参与了小肠移植后排斥反应过程,雷帕霉素既能抑制移植排斥反应,又可抑制ICAM-1的表达。 Objective:To study the relationship between the rejection and intracellular adhesion molecular (ICMI-1) of small bowel transplantation graft of rat and the action of rapamycin (RAPA) on it. Methods: Pseudooperated SD rats acted as control (group 1) and hetero-intestinal allotransplanted rats acted as model. Based on different treatment,these tested rats were divided into 3 groups:rejection (group 2),PARA 2 mg/kg(group 3) and PARA 4 mg/kg groups (group 4). All rats were killed and their small bowel transplantation graft were harvested for pathologic and ICMI-1 immunohistochemical tests on POD 1,3,5 and 7, respectively. Results: The pathological evidences of rejection were not detected in all groups on POD 1,3. There were episodes of 1?and 11?rejections in Group 2 on POD 5,7 respectively.On POD 7,the mild rejection happened in group 3, and no rejected trail existed in group 4. Only a little ICMI-1 expression in endothelium and lamina propria in control group,but the quantity of ICMI-1 expression in the same place reached a peak in rejection group on POD 5. There were significant differences in expression of lymphocyte and interstitial cell between the test and control groups. Conclusion; ICMI-1 takes part in the rejected process of small bowel transplantation. RAPA not only inhibits rejection,but also inhibits expression of ICMI-1 in small bowel transplantation.
